4、ccdingtotheacterofSsBHMTthatcanpromotesynthesisofproteinbyregulatingmethioninemetabolism.Metwassubstrateofadenosinemethionine(SAM)whichisdominatemethyldoninplant.Methylhavebeentakenpartinmetabolizingadjustingofplant.BHMT
5、wastransferredintoplanttoseekfactiontoplant.RNAwasextractedfromfreshpigliver.SsBHMTgenewasamplifiedbyRTPCR.AfterconfirmingthatitssequencewascrectbysequencingprokaryoticexpressionvectpET30aSsBHMTwasconstructedtransferredi
6、ntoE.coliER2566BL21(DE3).Theobjectiveproteinwasinducedpurified.BySDSPAGEelectrophesisanalysisitwasfoundthatthegenewasnotinducedinE.coliBL21(DE3)butmainlyexpressedinthefmofinsolubleinclusionbodyinER2566expressedalittleins
7、upernatant.ThentheconditionsofinducementincludingODvalueofbacilliinductiontemperatureIPTGconcentrationwereoptimized.Theexpressionofobjectiveproteinenhancedthanbefebutstillexpressedmainlyinthefmofinsolubleinclusionbody.Th
8、usinclusionbodydissolvedwithdenaturingagentwaspurtified.BySDSPAGEelectrophesisanalysistherewasaproteinbbetween45KDa66KDawhichsizeisthesameastheexpected.Meanwhiletheeukaryoticexpressionvectp3301SsBHMTcontainingCaMV35Sprom
9、oterwasconstructed.ThisvectwastransferredintoArabidopsisthalianabyflowerdipmethodofAgrobacteriummediated.25strainswerescreenedbysprayingrepeatedly0.5%(vv)herbicide.LeavesDNAof14strainswhichwereedfrom25transgenicstrainswa
10、sextracted.SsBHMTBARgeneof14strainsweretestedbyPCR.TheresultindicatedthatSsBHMThadbeentransferredintoArabidopsisthaliana.SsBHMTgenewasexpressedpurifiedtransferreditsplantexpressionvectintoArabidopsisthalianawhichprovides
