1、IGF1R 和 BRCA1 在乳腺良惡性腫瘤中的表達及其意義 中文摘要 背景和目的 乳腺癌是女性最常見的惡性腫瘤之一，其發(fā)病機理目前還不清楚。現(xiàn)有研究表明 BRCA1 和 IGF1R 與乳腺癌的發(fā)生、發(fā)展關系密切。BRCA1 是一種抑癌基因，具有負性調(diào)控細胞分裂周期和抑制腫瘤形成等作用。 BRCA1 與家族性乳腺癌的關系已經(jīng)明確， 在家族性乳腺癌中大約有 80%發(fā)生 BRCA1 基因突變。 但在散發(fā)性乳腺癌中還沒有發(fā)現(xiàn) BRCA1 基

2、因突變，不過有許多研究顯示 BRCA1 在散發(fā)性乳腺癌中會表現(xiàn)低表達或失表達，表明 BRCA1 對散發(fā)性乳腺癌的發(fā)生也有重要作用。IGF-IR 是一種酪氨酸激酶跨膜蛋白受體，與 IGFs 結合后可以介導 IGFs 的生理功能，促進細胞增殖，抑制細胞凋亡。文獻報道 IGF-IR 在乳腺腫瘤上呈現(xiàn)高表達，與乳腺癌的發(fā)生密切相關。IGF-IR 作為一種生長調(diào)控因子，它的基因表達受許多抑癌基因的調(diào)控，如 p53、BRCA1 等。最近有研究顯示

3、BRCA1 可抑制 IGF-IR 啟動子的活性，調(diào)節(jié) IGF-IR 蛋白的表達。目前，對 BRCA1 和 IGF1R 在乳腺癌中的表達及其關系還不太清楚。本實驗應用免疫組化 SABC 法檢測 BRCA1 和 IGF1R 在乳腺癌中的表達，同時分析它們與腫瘤類型、大小、分級及腋窩淋巴結轉(zhuǎn)移的關系，探討 BRCA1 和IGF1R 在乳腺癌診斷、治療和預后中的意義。 材料和方法 應用 SABC 免疫組化法檢測 30 例乳腺纖維腺瘤、42 例
